- 1. Medium filtration
Culture medium is an artificial nutrient for the growth and maintenance of microorganisms, plants and animal tissues. It is widely used in pharmaceutical processes such as biological fermentation and cell culture.
Because some media will destroy the effective components after high-pressure sterilization, it is necessary to eliminate bacteria and Mycoplasma through filtration to ensure sterility.
Different media contain different components. For media containing more proteins and sugars, multi-stage progressive filtration should be adopted to ensure the filtration capacity and the use efficiency of terminal sterilization filter element.
2. Buffer filtration
A buffer is a solution that resists pH changes when a small amount of acid or base is added. In biopharmaceutical, for example, buffer is used in the purification process of vaccines, blood products, monoclonal antibodies and so on. It is recommended to use a sterilizing filter to filter the buffer. If a pre filter is used, it can effectively protect the use of the terminal sterilizing filter. The reason why the buffer is filtered is that, on the one hand, it can protect the service life of the downstream chromatographic column and ultrafiltration membrane, on the other hand, it is to ensure the quality of the final vaccine.
3. Growth factor filtration
Growth factors are cytokines with the activity of stimulating cell growth. They are polypeptide substances that regulate cell growth and other functions by inducing binding with specific cell membrane receptors. At present, most growth factors are obtained through bioengineering technology, such as insulin.
4. Monoclonal antibody (mAb) is an antibody prepared by monoclonal technology. At present, large pharmaceutical enterprises have joined the field of mAb. In the process of McAb harvesting and purification, the target protein should be separated from impurities by purification process.
